Summary

It is a hybird promoter. One LacI binding site was added after TATA box of GAL1 promoter. TetR could bind to this promoter and thus repress downstrean gene transcription. Inducer (IPTG) could inhibit LacI protein and trigger gene transcription. (Ellis, Nature biotech, 2009, V27, 465-471)

培养条件 Strian grow in synthetic drop-out media without tryptophan (SD-TRP;Atkinson,Cell 113:597–607,2003) supplemented with inducer IPTG(10 mM) and 2% galactose for about 20 h to an optical density at 600 nm (OD600) of 1.00 at 30℃ with orbital shaking before measurement.
